Lentiviral Core Laboratory

The Lentiviral Laboratory provides lentiviral stocks for delivery and expression of transgenes in difficult to transduce primary cells and/or tissue culture. Services include: production of stable cell lines, lentiviral stocks for delivery and expression of transgenes, lentiviral delivery of siRNA or shRNA for gene silencing applications, backbone vectors for cDNA expression, small-scale virus production for gene transfer into cell lines, and high-titer virus production for gene transfer into primary cells and in vivo. Consultation for vector modalities, troubleshooting, and support documentation for grants is also available.

Molecular Genomics / DNA Oligo and Sequencing

DNA Sequencing services in the Molecular Genomics Core Facility are performed by GENEWIZ, Inc. Services include: Capillary DNA sequencing and oligonucleotide synthesis, evaluation of primarily genetic variations for association studies using single nucleotide polymorphisms (SNPs). Orders are placed directly on GENEWIZ’s website.

Molecular Genomics / Genomics

The Molecular Genomics Core Facility performs high throughput analyses of genetic (polymorphism/mutation) and epigenetic variations (DNA methylation) that underlie predisposition and progression to cancer. Services include: evaluation of inheritance of information on the basis of epigenetics. The use of genetically modified rats and mice as models for cancer and tumorigenesis is supported by genotyping mouse progeny in conjunction with the Transgenic/Knockout Core to follow genetic markers during mating. The Core utilizes expertise available within the Cancer Center for assist researchers withepidemiologic/population and DNA methylation studies.

Molecular Genomics / Methylation

The Genomics Core provides methods for evaluating the inheritance information on the basis of epigenetics. Services include: DNA extraction, Bisulfite conversion, MethyLight, CIMP Markers, Global DNA Methylation Analysis. Methylation-specific DNA extraction from cell lines, tissues, formalin-fixed paraffin-embedded tissues (either whole slide or microdissected tissues), as well as serum or plasma are compatible with DNA methylation assays, providing there is sufficient amounts of DNA in each sample.

Molecular Genomics/Microarray

The USC Norris Comprehensive Cancer Center Microarray Core is located at the CHLA pediatric facility. Services include: gene expression profiling with a variety of arrays, including human, mouse and rat; mutation analyses (p53); HIV, yeast, and bacterial genome analyses; and SNP analyses. RNA or DNA is extracted, purified, labeled, and in the case of expression chips, converted into double stranded cDNA, prior to cRNA, labeling and fragmentation.

Norris Library Bioinformatics Support

Bioinformatics support is provided by the Norris library to faculty, students and researchers. Services include: statistical analysis of high-throughput data and DNA/protein sequence analysis, comprehensive functional analysis and advanced literature and data search and mining, as well as workshops and training of available software tools.

UPC Genome & Cytometry Core

The Genome & Cytometry Core is located on the UPC campus. Equipment and services include: High-throughput next-generation sequencing on the Illumina HiSeq 2500 and the Ion Torrent PGM platforms. The HiSeq 2500 system can be used in Rapid Run mode (equivalent to MySeq) or High-output mode. The Core also provides optional library construction services and use of our high-throughput Beckman Coulter Biomek® 3000 Automated Workstation as well as access to the BD Influx Flow Cytometer & Cell Sorter.

Vector Core

The Vector Core provides researchers with state of the art technology, facilities, support staff and services and a broad range of resources. Services include: assistance in the design and construction viral vectors and viral particles for gene-modified in vitro/in vivo studies. The core also provides consulting services to identify researcher gene transfer needs, determine the best way to produce gene-modified cells, and to assist in experimental troubleshooting. Equipment includes: Hollow fiber tangential flow filtration (TFF) columns (Spectrum Labs) to filter and concentrate lentiviral vectors up to 100 fold with high recovery (>90%).
